OT: Employer/Employee Reference Question

If anyone has some good advice, it would be appreciated. My wife works for a small family-owned business, loves her job, however one of the other ladies, who doesn't get along with one of the bosses, is looking for another job and wants to use my wife as a reference. Should my wife say yes and if so, should she inform her bosses?
